Measurement of Sodium and Potassium in Cheese 1 Dried milk is dissolved in warm water. The test solutions and reference solutions are measured directly following dilution with deionised distilled water. Procedure Thoroughly clean and rinse all glassware with deionised distilled water prior to use. It is recommended that polyethylene is used to ensure that sodium does not plate out onto glassware surfaces. If only glass is available, the sodium determination was carried out rapidly. 1.25grams of cheese was accurately weighed out on a 5-figure analytical balance and recorded for backtracking the concentration to the sample size. The sample was then added to a crucible of appropriate size and 10ml of ~30% sulphuric acid. It was ensured that the crucible was less than half full of both the sample and acid added to avoid boiling sample being spilled as it was heated. The oven was heated to 600 degrees Celsius, and the crucible was weighed on the 5-figure analytical balance. The crucible was then added to the oven and left to ash over the course of 2 hours. The sample was then reweighed and placed back into the oven for 5 minutes. The crucible was then weighed again, if the weight of the crucible has not changed from the previous weight, then the sample was finished with the ashing process, if it had significantly changed the process was repeated until all combustible content of the sample had been removed. The ashed sample was then dissolved in 54ml of DI water with 1ml of 10% HCl. This was then filtered through a funnel and Whatman n.5 filter paper into a 100ml polyethylene volumetric flask and made up to 100ml with DI water. A minimum of 5 standards were produced with a concentration range of 0-100mg/l potassium and sodium, to each standard 10ml of ~30% sulphuric acid was added and 1ml 10% of HCl was added. (If the sample contained a higher concentration than these standards, the sample solution was further diluted to bring the concentration under 100mg/l.) The standard was calibrated starting with the lowest concentration standard, working up the concentration gradient over the 5 standards. The results will not be reliable if this solution contains insoluble particles. If insoluble particulate matter is present in the sample, filter the sample through Whatman No.1 filter paper. The filter paper should be weighed on an analytical balance prior to filtration, and then the solution filtered and the paper dried in an oven at low temperature then reweighed and the difference calculated between the pre and post filter paper. This should then be accounted for when calculating concentrations by weight. Preparation of Standard Graph Set the flame photometer in accordance to MultiPoint/Single Ion Calibration found on page 24 of the BWB Technologies Installation and Operation Manual, to measure potassium emission. Nebulise the working standard solutions and adjust the controls until steady zero and maximum readings are obtained. Measurement of Potassium and Sodium in Meat 1 Several different techniques can be used for sample digestion and extraction of sodium and/or potassium: oven ashing, perchloric acid ashing, extraction by boiling and acidification with HNO3 as well as extraction by homogenisation in 2% trichloroacetic acid (TCA). Results of this investigation indicated that the TCA extraction method was repeatable and simple. The recovery of known amounts of potassium showed the method to be accurate. The results for sodium showed more variability and gave a value lower than expected by about 5%. The amount of potassium found in pork and lamb ranged between 1750 and 3070ppm and the amount of sodium to be between 350 and 700 ppm. Procedure All solutions are prepared using deionised distilled water unless prompted otherwise. A sample weighing between 1.5 and 3.5grams of tissue is homogenised in 150ml of 2% trichloracetic acid (TCA) and stored overnight at 5ºC in polypropylene containers to avoid sodium plating out onto glassware. The homogenised sample is filtered through Whatman number 40 filter paper into clean polyethylene bottles. A 5ml aliquot of the sample is made up to 15ml with 2% TCA. This solution is then used for sodium and potassium determination. Standards are then made for both sodium and potassium around the expected result. If unsure about the range in which the sample will be read at, the BWB Technology offers a single calibration point analysis. This can then be used to estimate an expected range of results and calibrated after this initial estimation of sample. Standards should be aspirated in order of lowest to highest to avoid previously ran standards contaminating the further calibration standards. It should also be noted that it is of vital importance that further sample dilution is required if the concentration is coming out at over 100ppm. Whilst the BWB Flame Photometer is able to detect ion concentrations over 100ppm, interference begins to become significant in the readings over 100ppm as well as matter being deposited onto the nebuliser array which can be released in later aspirations increasing signal noise and data stability. When Calibrating, all standard solutions should be made up with 2% TCA solution as well as a solution of 2% TCA ran as a blank to reduce chemical interferences. The blank can be run when prompted on the full range of BWB flame photometers. Chemical interference for sodium readings can be reduced by neutralising the solution with a few drops of base solution, however it should be taken into consideration that the base selected for neutralisation should not contain any ions that can contaminate readings. Principle Potassium is extracted from soil with ammonium nitrate. Apparatus Bottles - 125 ml, wide mouth, screw cap. Scales - accurate to 1 DP. Shaking machine with platform - approx. 275 strokes per min, length of travel 25 mm. Reagents Ammonium nitrate, M - Dissolve 400 g of ammonium nitrate in water and dilute to 5 l. Standardization is unnecessary. Method Grind soil and pass through a 2 mm mesh sieve. Transfer 20 g of air dried soil into a bottle and top up to 50 ml of M ammonium nitrate. Cap the bottle and shake on the shaking machine for 30 mins. Filter through a 125 mm Whatman No. 2 filter paper and retain the filtered extract (this sample may also be used for the determination of magnesium; and sodium if carried out immediately or the bottle is stored in a polythene bottle). Carry out a blank determination with ammonium nitrate solution. Determination The concentration of potassium in the extract is determined by flame photometry; significant interference by other elements does not occur. Reagents Potassium working standard solutions, 0-50µg/ml of potassium - Prepare solutions containing 0, 10, 20, 30, 40 and 50 µg/ml of potassium in 1M ammonium nitrate. Preparation of Standard Graph Set the flame photometer in accordance to MultiPoint/Single Ion Calibration found on page 24 of the BWB Technologies Installation and Operation Manual, to measure potassium emission. Nebulize the potassium working standard solutions containing 0 and 50 µg/ml of potassium and adjust the controls until steady zero and maximum readings are obtained. Nebulize the intermediate working standard solutions and construct a graph relating meter readings to µg/ml of potassium in all the standard solutions. Examination of Extract Adjust the flame photometer until the steady zero and maximum readings are obtained with potassium working standard solutions containing 0 and 50 µg/ml of potassium. Nebulize the extract and note the meter reading. Calculation of Results Read from the standard graph the number of µg of potassium equivalent to the meter readings of the sample and the blank determinations. Multiply the difference by 5. The result gives the mg/l of extractable potassium in the soil. Appendix Potassium stock standard solution, 1 mg/ml of potassium - Dry potassium nitrate at 102°C for 1 hr and cool in a desiccator. Dissolve 1.293 g of the dried salt in water and add 1 ml of hydrochloric acid, approx. 36% m/m HCl to reduce the pH and aid dissolution of the sample. Dilute to 500 ml and add 1 drop of toluene. A simplified menu structure and enhanced Li detection make it a welcome addition to the BWB family. The ability to calibrate and measure Sodium, Potassium, and Lithium at the same time with the same dilution (from 1:6 to 1:100) streamlines throughput in many laboratories. We have retained the ability to enter and store two user-defined calibration curves should the user wish to readily measure differing sample types. When connected to a PC with the BWB FP-PC app, the user can create .pdf files, .csv files, data presentations and reports that include all the key analysis data. The FP-PC Software also allows setup of the Automatic Fluid Handling System, AFHS, originally designed to work specifically with the BIO-943 unit. The software enables you to view and edit the calibration curves. The intuitive user-friendly operation of the BWB BIO 943 spans all languages and appeals to all levels of users. The BWB BIO 943 is the must-have instrument in today’s research laboratory. For sites that do not allow PCs or data acquisition in the test lab, the Printer provides the “hard copy” required for QA and QC purposes.
